Warcraft 3 does not help a whole lot, other than that you might recognize some models that were carefully "re-imagined"/ripped off.
Basically you control a hero. You run around in one of the three lanes, killing the creeps that march repeatedly to their death to gain gold and experience (don't autoattack them constantly though, this will quickly get you hated by everyone. Just last-hit, meaning that you score the final blow to kill off the creep). When you level up, you gain abilities and shit that you can use to fuck over your team or sometimes, if you're lucky, kill off an enemy hero.
You destroy the towers in each lane, get to your opponent's base and bash down their main structure. Easy as that!
Oh, and you should also buy items. Preferably those that work for your character. There's one item shop in your spawn, one to the side of each sidelane shop, and also secret shops for certain items. When you open up the shop window, you'll see a recommended build. Just buy whatever is there!
IT'S ALL VERY SIMPLE REALLY no seriously you'll have to play like 50 games before you even gain basic understanding of it. Have fun dying. Repeatedly. And getting blamed for every loss every game. <3
Seriously, though. It's a super fun game, once you get into it.
Pro tip: When you see an enemy hero, click on him and read the abilities that he has. This assures that you at least know WHAT'S killing you. Even if you can't stop it.
OH AND FOR THE LOVE OF THRALL: Don't fucking click on your abilities or items to use them. Hotkeys are essential, and if you don't use them you'll be more wrecked than the 10-year old I raped last week. Seriously, she still can't walk.
